Introduction to immunodeficient animals


Concept: Immunodeficiency animals are animals that have defects in one or more of the components of their immune system, either because of an innate genetic mutation or because of artificial methods.


Classification by cause:


Congenital immune deficiency animals: also known as spontaneous immune deficiency animals, refers to the naturally occurring immune deficiency diseases of animals and the animals with immune deficiency or low function acquired through directional breeding.


Acquired immune deficiency animal: also known as induced immune deficiency animal, refers to the animal whose immune function is deficient or low by artificial intervention, mainly genetically modified immune deficiency animal. Such as AIDS model, immunosuppression model.


Classification by degree of immune deficiency:it can be divided into T lymphocyte deficiency, B lymphocyte deficiency, NK cell deficiency and combined immune deficiency animals.


Immunodeficient animals application areas:


1. Oncology research: tumor growth, tumor metastasis, tumor recurrence and anti-tumor drug screening.

2. Immune system research: immunodeficient animals can be used in the study of immune system such as T cells, B cells, NK cells and immune-related diseases due to their immune function defects.

3. Liver research: mainly refers to liver regeneration function, drug metabolism and liver related diseases.

4. Infectious diseases: Because defective animals are sensitive to environmental bacteria and viruses, they are good animal models for studying viral and bacterial infections.


Immunodeficient dog model:


RAG2 gene editing dogs -- RAG2 gene deletion blocks T and B cell differentiation earlier and more completely, resulting in a deficiency of T and B cells, but normal NK cell levels.

PRKDC/IL-2RG gene editing dog -- No mature T, B and functional NK cells, does not reject allotransplantation and xenotransplantation.

RAG2/PRKDC/IL-2RG gene editing dog -- No mature T cells, B cells and functional NK cells, does not reject allotransplantation and xenotransplantation.
