Global cerebral ischemia Model of beagle
Ischemic cerebrovascular disease is the second leading cause of human death after ischemic heart disease.
Experimental animal: Beagle, 12-15 kg, male and female, about 1 year old, good health condition.
Model making method:
1.The carotid artery and deep vertebral artery were separated from the neck of the experimental dog along both sides of the trachea.
2.Mechanical control of respiration and tidal volume regulation by end-expiratory partial pressure of carbon dioxide.
3.0.02% sodium nitroprusside was dropped intravenously to reduce the average arterial pressure and maintain it at 6-7kpa.
4.The bilateral vertebral arteries and bilateral carotid arteries were clamped for 5 minutes, and then the clamps were loosened and sodium nitrate was stopped.
5.Average arterial pressure was maintained above 11kaP by intravenous drip of epinephrine.
6.After ischemia, 5% sodium bicarbonate 5mg/kg, low molecular dextran 5mL /kg, dexamethasone 5mg were dropped in 2 hours at a constant speed.
7.Skin closure: suture the incision layer by layer, first close the muscle layer and fascia layer with a 4-0 line of round needle, and then close the skin layer with Angle needle. Iodophor is used to disinfect the wound and a sterile dressing is applied. Intramuscular injection of ceftriaxone sodium (5mg/kg) for anti-staining, intramuscular injection of 0.5ml tongliding, put it into the nursing cage for observation.
Model identification:
1. Hemorheological indicators: plasma specific viscosity and hematocrit.
2. Number of animals survived after 24 hours.
3. Blood glucose indicators
Application:
It can be used to study the changes of cerebral blood flow and neurotransmitters after cerebral ischemia and neuroprotection from hypothermia. Combined with this method, vascular dementia model can also be made, such as high-fat feeding plus two-vessel block, two-vessel block plus coccygeal bloodletting, two-vessel block plus nitropenol.
