Canine model of infertility disease (Chronic salpingitis)

Clinically, chronic salpingitis is a common cause of infertility, which can cause fallopian tube obstruction and reduced peristalsis ability due to inflammation and lead to infertility, including changes in organ quality and function. This model study shows that it is basically consistent with clinical practice.

Experimental animals: 30 female domestic dogs, weight 10kg, 6-8 months old.

Model making method:

The anterior abdominal wall was surgically cut and both fallopian tubes were exposed. The mixed bacteria containing Escherichia coli, STaphylococcus aureus and Streptococcus were diluted with sterile normal saline at a ratio of 2:1:1. The concentration was 3*109/ mL and 0.5ml with a 1ml syringe, and then the fallopian tubes were injected from both sides of the dog in the direction of fallopian tube and ovary.

Model identification:

Naked eye observation: observe the shape of the fallopian tube, color change, and the surrounding tissue adhesion condition, whether thickening or thinning, elastic changes, and whether there is water, suppuration, etc. Observation of pathological changes: after the naked eye observation of the sacrificed animals, the fallopian tubes were accurately taken and fixed in 10% neutral formaldehyde solution, routine sections,HE staining, and pathological changes were observed under light microscope. The fallopian tubes were fixed with 4% glutaraldehyde phosphate buffer, then fixed with 1% Russian acid, dehydrated with acetone and embedded with oxidizing resin. Ultrathin sections were observed under transmission electron microscope. After modeling, adult male dogs of the same species were put in to observe the pregnancy situation.

Application: To study the etiology of pyometriasis in dogs.
